The call

The vaccine revenue that funded BioNTech is fading fast, and its cancer bet has not yet produced a single approved drug or its founders' successors.

BioNTech made huge profits from its COVID vaccine (Comirnaty) but sales are now falling fast. The company is betting on new cancer medicines, especially a drug called pumitamig that is still in testing. The challenge is that larger rivals like Merck, Roche, and AstraZeneca already dominate cancer treatment with proven drugs and stronger sales teams. Meanwhile, the company's founders who designed its science are leaving by the end of 2026 to start a competing company, which could weaken BioNTech's direction and take some of its technology with them. The one thing that could change everything is if pumitamig succeeds in its final-stage trials and if BioNTech hires a strong new leadership team to replace the founders.

Why
01
AI pressure: AlphaFold/Isomorphic Labs, Chai Discovery. AI speeds up early drug design and target discovery but cannot run trials or take on regulatory liability, so it sits inside the pipeline, not against it. Could AI do the core job? Unlikely.
02
Bigger players: Merck (Keytruda), Roche/Genentech, AstraZeneca, Bristol Myers Squibb. These companies already sell approved cancer drugs with huge sales forces, while BioNTech's oncology candidates are still in trials with no product on the market. Could a giant copy it? No, they are blocked.
03
What protects it. Patent-protected pipeline data, €17.2B cash to fund trials for years, and regulatory approvals already in hand for its vaccine platform.
What would change this call
Pumitamig Phase 3 ROSETTA Lung-01 readout reported through late 2026/2027. Positive data would validate the oncology pivot; failure would remove the main growth thesis.
Announcement of new CEO/CMO replacing Sahin/Tureci by end-2026. Credible leadership signals continuity; weak or delayed picks signal execution risk.
FY2026 revenue lands within or below the guided EUR2.0-2.3B range. Confirms whether the Comirnaty revenue decline is stabilizing or still accelerating.
Key numbers
  • Q1 2026 revenue €118.1M, down 35% YoY (2026-03-31)
  • €17.2B cash and marketable securities (2025-12-31)
  • FY2026 revenue guidance €2.0B to €2.3B (2026-05-05)
Named risks
  • Founders Sahin and Tureci departing by end of 2026 to launch a competing mRNA company, taking mRNA rights. Severity: high
  • Core Comirnaty revenue in freefall, down 35% YoY in Q1 2026 with no near-term replacement revenue. Severity: high
  • Oncology pipeline is pre-revenue and binary on Phase 3 readouts including ROSETTA Lung-01. Severity: high
  • Manufacturing site closures and ~1,860 job cuts through 2027 signal capacity retreat. Severity: medium
  • Larger oncology incumbents (Merck, Roche, BMS) have far greater commercial distribution scale. Severity: medium

Questions people ask
Is BioNTech profitable in 2026?
No. BioNTech posted a Q1 2026 net loss of 531.9 million euros as Comirnaty revenue fell 35% year over year to 118.1 million euros. FY2025 net loss was about 1.14 billion euros, and FY2026 revenue guidance of 2.0 to 2.3 billion euros is well below 2025's 2.87 billion euros.
Is BioNTech laying people off?
Yes. In May 2026 BioNTech announced closures of manufacturing sites in Idar-Oberstein, Marburg, and Tubingen in Germany plus Singapore, affecting up to about 1,860 positions through end of 2027. Separately, roughly 820 jobs are being cut at CureVac following BioNTech's December 2025 acquisition.
Could AI replace BioNTech's scientists?
Unlikely soon. AI tools like AlphaFold and Chai Discovery accelerate early target identification and antibody design, but cannot run Phase 3 trials, bear regulatory liability, or manufacture at scale. BioNTech owns its own AI unit, InstaDeep, acquired in 2023 for about 362 million pounds, embedding AI as an internal tool rather than a replacement.
Who owns BioNTech?
BioNTech SE trades on Nasdaq as BNTX, but the Strungmann brothers, Thomas and Andreas, remain the largest shareholders via AT Impf and Athos, holding roughly 40% as of BioNTech's 2025 annual report. Co-founder Ugur Sahin also holds a meaningful personal stake, though he is departing the company by end of 2026.
Who is BioNTech's biggest competitor?
Moderna is the closest direct mRNA-platform peer, also pivoting from collapsed COVID vaccine revenue toward oncology with Merck. In oncology specifically, BioNTech's pumitamig program competes against entrenched giants Merck, Roche/Genentech, and Bristol Myers Squibb, the latter now also a co-development partner on pumitamig itself.
What happens to BioNTech's mRNA technology when the founders leave?
Ugur Sahin and Ozlem Tureci plan to depart BioNTech by end of 2026 to found an independent mRNA-focused company, taking a minority stake and certain mRNA rights or technology with them, according to BioNTech's March 10, 2026 announcement that sent BNTX shares down as much as 17 to 22 percent.
