The call

Enhertu keeps growing fast and the chemistry lead holds, but a manufacturing miscalculation and rising Chinese ADC competitors are the first real cracks to watch.

Daiichi Sankyo makes Enhertu, a cancer drug that attaches medicine directly to tumor cells using a special chemistry process the company invented. It is hard to copy right now because the company owns the recipe, has approval in 90+ countries, and locked in deals with bigger drugmakers AstraZeneca and Merck to sell it. However, Pfizer (which bought the company Seagen) and Chinese competitors like Kelun-Botai and Hengrui are building similar drugs in the same space, and Daiichi Sankyo just admitted it overestimated factory capacity by roughly a billion dollars in 2025, which raises questions about whether the company can execute flawlessly. The biggest wildcard is whether Enhertu keeps growing fast enough to offset the company's reliance on just this one drug and the looming patent expiration around 2030 to 2035.

Why
01
AI pressure: AI-driven drug design labs (Insilico Medicine, Xaira Therapeutics). AI-assisted molecule design could speed rivals' ADC discovery, but has not replaced Daiichi Sankyo's DXd linker chemistry yet. Could AI do the core job? Unlikely.
02
Bigger players: Pfizer/Seagen and Roche ADC portfolios, plus Chinese entrants Kelun-Botai and Hengrui. Kelun-Botai's sacituzumab tirumotecan is FDA-approved and Hengrui's rezetecan is advancing, both competing directly in the same Trop-2/HER2 ADC space. Could a giant copy it? No, they are blocked.
03
What protects it. Proprietary DXd linker-payload chemistry, approvals in 90+ countries, and locked global distribution deals with AstraZeneca and Merck.
What would change this call
FDA PDUFA decision on ifinatamab deruxtecan due October 10, 2026. Approval would extend the DXd pipeline; a delay or rejection would weaken pipeline de-risking.
Company discloses resolved capacity/production plan within about 6 months of May 2026. Shows whether the $950M forecasting error was a one-off or reveals ongoing execution risk.
Kelun-Botai or Hengrui ADC sales/label expansions reported in US or EU markets within 12 months. Would show Chinese entrants converting approval into real revenue share against Daiichi Sankyo.

Questions people ask
Is Daiichi Sankyo profitable in FY2025?
Yes. For the fiscal year ended March 31, 2026, Daiichi Sankyo reported consolidated revenue of JPY 2,123.0 billion (+12.6% YoY) and core operating profit of JPY 360.0 billion (+15.1% YoY), despite absorbing a ~JPY 149.4 billion one-time manufacturing charge, per its May 11, 2026 results.
Why did Daiichi Sankyo take a $950 million loss?
Daiichi Sankyo overbuilt ADC manufacturing capacity relative to revised demand forecasts, resulting in a ~JPY 149.4 billion ($950M) extraordinary loss disclosed May 8, 2026, including JPY 75.7 billion in CMO compensation payments and an Odawara plant impairment charge.
Who is Daiichi Sankyo's biggest competitor in ADCs?
Pfizer/Seagen has been its longest-running rival via ADC patent litigation, though Daiichi won a Federal Circuit appeal reversing a $41.8M verdict in December 2025. Newer threats include FDA-approved Chinese ADC entrants Kelun-Botai and Hengrui in overlapping Trop-2/HER2 target space.
Could AI replace Daiichi Sankyo's drug development work?
No. AI is being used as an accelerant, not a replacement: Daiichi Sankyo signed AI partnerships with FRONTEO (Aug 2025), Lunit (Dec 2025), BostonGene (Feb 2026), and Tempus AI (March 2026) for toxicology and biomarker work, but its regulatory approvals and manufacturing moat remain unreplicable by AI.
Is Daiichi Sankyo hiring or cutting jobs?
Daiichi Sankyo is growing headcount. Company-reported group-wide employees were 19,765 as of March 31, 2025, up 1,039 (+5.55%) year over year, driven by global oncology commercial and R&D expansion, per its official sustainability data sheet.
Who owns Daiichi Sankyo?
Daiichi Sankyo is an independent public company with no parent, trading on the Tokyo Stock Exchange Prime Market under ticker 4568, with a market capitalization of approximately JPY 5.02 trillion as of June 2026.
