Sarepta Therapeutics
Gene therapy maker selling ELEVIDYS and PMO drugs for Duchenne muscular dystrophy.
Sarepta still leads DMD treatment with approved drugs and Roche's backing, but three ELEVIDYS deaths, an EU rejection, and rival gene therapies coming through 2026-2027 leave its lead exposed.
Sarepta makes four FDA-approved drugs for a rare muscle disease called DMD, bringing in about $1.86 billion per year. But in 2025, three patients died after receiving its main new therapy ELEVIDYS, which forced the company to add a serious warning label, cut its staff by more than a third, and lower its 2026 revenue forecast from $1.86B to $1.2-1.4B. The company has real advantages: FDA approval, specialized manufacturing, and a partnership with the giant drugmaker Roche. However, these advantages are being challenged by at least three competitor companies working on similar DMD treatments, including Dyne Therapeutics, which is expected to get FDA approval for its own DMD drug in January 2027 and could take sales from Sarepta's older pill-based treatment. The key question is whether Dyne's new competitor drug will be seen as safer or more effective, which would carve into Sarepta's existing customer base.
ENDEAVOR Cohort 8 sirolimus pretreatment data readout. Safe results could restore non-ambulatory ELEVIDYS access lost after 2025 deaths.
Outcome of Roche's new global Phase 3 Elevidys trial for EU resubmission. Success or failure decides whether Sarepta regains European market access ceded after CHMP rejection.

| REGENXBIO | gaining on | RGX-202 gene therapy filing expected mid-2026, per Feb 2026 report |
| Solid Biosciences | contesting | SGT-003 in Phase 1/2 as of early 2026, advancing rival DMD gene therapy |
| EU regulators/CHMP | losing to | ELEVIDYS marketing application rejected in EU in 2025, forcing new Phase 3 trial (Apr 2026) |
- Is Sarepta Therapeutics profitable in 2026?
- No. Sarepta guided 2026 net product revenue to just $1.2B-$1.4B, down from $1.864B in 2025, after a safety crisis cut ELEVIDYS sales; a GAAP net loss was reported for 2025 despite $1.86B revenue.
- Did Sarepta Therapeutics have layoffs?
- Yes. In July 2025 Sarepta cut about 493 confirmed jobs, roughly 36% of staff, to save an estimated $400M annually starting 2026, shrinking headcount from 1,372 (end 2024) to about 835 by July 2026.
- Is Sarepta's ELEVIDYS gene therapy safe?
- It carries serious risk. Three patients died of acute liver failure in 2025, prompting a July 2025 FDA black-box warning and removal of the non-ambulatory indication; a new sirolimus-pretreatment cohort is testing whether access can be safely restored.
- Who is Sarepta Therapeutics' biggest competitor?
- Dyne Therapeutics is the most direct threat: its DYNE-251 has FDA priority review with a PDUFA date of January 21, 2027, targeting the same exon-51 Duchenne population as Sarepta's $965.6M-a-year Exondys 51 (2025 revenue).
- Who owns Sarepta Therapeutics?
- Sarepta is a publicly traded Nasdaq company (SRPT) with no parent; market cap was about $1.67-1.68B as of July 23, 2026. Roche has held its ex-US ELEVIDYS commercialization rights since a December 2019 licensing deal.
- Could AI replace Sarepta Therapeutics?
- Unlikely. Sarepta's value rests on FDA biologics licenses for four approved DMD drugs, AAV/oligonucleotide manufacturing know-how, and its Roche distribution partnership, none of which AI models can replicate or absorb as of this brief (2026-07-24).
